This item is found at Dead Tree, a location inside Dead Roots which requires the Dead Roots Key to access.
The Glowstone Backpack can be found hidden behind the west railing of the staircase across from the Ornate Chest.
To obtain the Dead Roots Key, the player must find all four notes (not just 3) during Purifier.
Note: the backpack can still be obtained after the completion of Purifier if you collected 4 keys during the quest.
